Biosensing devices based on novel biochips and tuning of laser diodes

Obiettivo



The analysis of interactions involved in biomolecular complexes is of fundamental importance for the understanding of biological systems. Frequently used tools for these studies are immunoassays as ELISA and RIA. Surface plasmon resonance (SPR) is a relatively new tool for functional studies of biomolecular complexes. In a study, funded by an Exporatory Award of the Biotechnologies programme of the EC, that we just completed, we investigated a new effect in SPR use: Ordinary semiconductor laser diodes, which are sold in industry for as little as about 10 ECU each, can be frequency tuned with extreme precision. This can be applied also in SPR biosensing with very high accuracy at very little costs .
In the proposed project a device based on this novel principle and combining this development in a mutual "symbiotic" approach with research on novel sensing layers will be developed. They will consist of gold supports, hydrogels of synthetic macromolecules that are functionalized with biomolecules (e.g. recombinant proteins) or semisynthetic receptors (e.g. based on cyclodextrins). Novel stimulus responsive films, using temperature dependent conformation changes of hydrogels and novel SPR signal enhancement structures will be synthesized and evaluated.
We apply for a Shared-Cost Measure under the Biotechnologies programme in 6.2.2.Main proposer is MiNT, a Germany-based SME, that has experience in combination of surface science, optics and biochemistry and experience in project management.
The development of the SPR biosensor requires a multi-disciplinary team with experience in different fields ranging from optoelectronics, over chemistry to biology. Each of the proposers is expert in its field and its participation is essential for the success of the project.
- MiNT is a German SME founded by the physicist Dr. G. Brink. This SME cares for all the coordination among the partners, will use its knowledge of ultrathin films and surface treatment and will take care of the exploitation of the project results.
- CYTOS, biotechnology SME and spin-off of the biotechnologies faculty at theETH Zurich, is directed by Dr. W. Renner. It will be responsible for the biomolecules used in the project and for all biotechnological aspects. - a Finnish group, directed by Dr. T. Sadowski (VTT Chemical TechnologyInstitute, Tampere) will be responsible for the development of theSPR-device.
- a French group directed by Dr. L. Jullien (Departement de chimie, ENS),will study the effect of SPR enhancing NIR dyes and will prepare the synthetic receptor molecules.
- a Belgian group, directed by Prof. A. Laschewsky (Departement de Chimie, Universite Catholique de Louvain La Neuve) will be responsible for the preparation of the ultrathin films. Prof. Ringsdorf from Mainz and Londonwill assist the project with his long-time scientific experience.
Very often technology that was invented and developed by European researchers has reached mass production and consumer markets only after exploitation in other countries as Japan or the USA. We want not to let this happen in the case of affinity sensing. We believe, that cheap and smart, mass market products can be developed in Europe as well. At the same time we will enter new fields of scientific discovery in a very interdisciplinary approach of researchers from very different fields and different types of laboratories.
