Ultra-sensitive multianalyte immunoassay based on dna-senor

Objectives and content
Whilst radio immunoassay (RIA) and enzyme-linked
immunosorbent assay (ELISA) are well-established
analytical methods in clinical laboratory, there is a
need of ultra-sensitive multianalyte immunoassays for
sensitive and reproducible quantification of analytes in
many areas that are currently problematic, including
diagnostic medicine and the standardisation of
biological. The use of immuno-PCR in combination with a
sequence-selective DNA-sensor based on micro-technology
could provide the basis for the simultaneous detection of
many analytes at low cost and at sensitivities higher
than those of existing antigen detection systems.
In a feasibility study suitable intercalators and
electrode materials have been selected and double
stranded DNA could be detected using amperometric method.
In principle, therefore, this configuration is well
suited for use as DNA-sensor.
The purpose of this proposal is thus to develop a
prototype for the simultaneous determination of two
analytes based on novel sequence-selective DNA-sensor
using micro-technology. To achieve this objective, the
proposed research project will include:
development of immuno-PCR for the simultaneous
determination of two analytes,
design and production of addressable electrode arrays
in the microfabrication range for the DNA-sensor,
amperometric or potentiometric DNA-sensor for the
determination of two different DNA as quantification step
of the immuno-PCR,
development of the performance of the complete
prototype system.
This new technology will improve the quality of European
industry in the area of new functionalised materials and
new analytically methods since new innovative
technologies and a totally new product will be developed
and patented for the future world market which will be of
considerable added value. It should be stressed that in
this project three SMEs (Inventus BioTec GmbH & Co - KG
(Germany), Biosensores, s.l (Spain) and PharmaGen SA. -
(Spain) are included and they could build an efficient
transitional network of complementary SMEs for the
development of novel technologies for the diagnostic
medicine.
