Construction of micro-satellite markers maps for Atlantic salmon (Salmo salar), rainbow trout (Oncorhynchus mykiss) and brown trout (Salmo trutta)

Within this project, 954 micro-satellite PCR assays have been described and fully characterized for salmonids and 307 additional PCR assays have been partially characterized. For Atlantic salmon a total of 639 microsatellite loci have been identified (555 from Atlantic salmon, 57 from rainbow trout and 27 from brown trout) and partially characterized and so far 447 (363 from Atlantic salmon, 57 from rainbow trout and 27 from brown trout) have been fully characterized. 300 markers (299 DNA-markers + sex) were genotyped in the reference families and subsequently mapped by linkage-analysis resulting in the identification of 35 linkage-groups but with 34 markers still unassigned to a linkage-group. For rainbow trout a total of 407 micro-satellite loci have been identified (215 from rainbow trout, 165 from Atlantic salmon and 27 from brown trout) and 292 (173 from rainbow trout, 92 from Atlantic salmon and 27 from brown trout) have been fully characterized. 299 markers and sex were genotyped in reference families. Linkage-analysis resulted in the identification of 31 linkage groups including 269 loci but with 31 markers still unassigned to a linkage-group. For brown trout a total of 215 micro-satellite loci have been fully characterized (27 from brown trout, 50 from rainbow trout and 138 from Atlantic salmon). Linkage analysis resulted in the identification of 35 linkage-groups including 222 markers but with 11 markers still unassigned to a linkage-group. All sequences are submitted to GenBank and all information on the markers and the marker maps will be made available to all interested through publication in international journals and will after publication also be available from a WEB site. The results include sequences of clones containing micro-satellites, information on primer sequences, PCR-conditions and information on polymorphism in the three species Atlantic salmon, rainbow trout and brown trout. The results also include information on how suitable the markers are for genotyping both with radioactivity and with fluorescence. Finally the results include information on the genetic maps for all three species.
