Objectif Endoglin is a homodimeric membrane glycoprotein, highly expressed in endothelial cells and macrophages, and with capacity to bind transforming growth factor-Beta (TGF-Beta). Endoglin is a component of the membrane TGF-Beta receptor system, although its specific role in the TGF-Beta induced cellular responses is not known. To study the functional role, we will analyze the TGF-Beta responses of stable transfectants in human U-937 cells expressing two different endoglin isoforms (L-endoglin and S-endoglin) with distinct cytoplasmic tails. U-937 cells are responsive to TGF-Beta, as previously studied in our laboratory, and we will analyze the cellular proliferation, migration, aggregation, fibro- nectin synthesis, upregulation of membrane adhesion molecules, levels of c-myc, c-fos and c-jun mRNA, as well as the TGF-Beta binding capacity of the different membrane receptors.
