Characterization of the mechanisms of transcriptional activation by peroxisome proliferator-activated receptor

Obiettivo



Peroxisome proliferator-activated receptor (PPAR) binds to specific DNA sequences and thereby activates gene transcription.
The aim of this project is the understanding of molecular mechanisms of gene regulation by PPAR. The PPAR mediated gene activation, which has been studied in the host institute intensively, serves as a suitable model to understand general principles in gene regulation.
In the host group different PPAR interacting proteins have been identified. These are the basis for different approaches to reveal PPAR mediated gene activation.
Full-length cDNA of still identified PPAR interacting proteins will be obtained by phage screening and their biological role will be characterized in cell culture DNA-transfection experiments.
To identify their interacting partners, proposing PPAR interacting factors will be used in vivo in a yeast two hybrid system and in a new in vitro system, which has to be adapted for phage screening.
Positive interacting factors will be analysed in an in vitro transcription assay and in cell culture and thereby, the general validity of PPAR mediated gene regulation for other hormone receptors will be tested.
