The project presented will provide new data to understand the chronotropic acclimation response at low temperatures and investigate the mechanisms proposed for the inotropic response in a teleost species: the Atlantic cod, Gadus morhua. The experimental approach will also contemplate the length of the acclimation time because this is one of the variables that lead to different conclusions in the previous literature on the subject In experiment I atrial muscarinic and p-adrenoreceptors will be quantified using a slide/mircopunche technique at two different acclimation temperatures (low=5QC and control=lOrC). Since the binding to muscarinic and ,B-adrenoreceptors display an antagonistic action their balance should lead to a compensated increase in heart rate coincident with the observed Q10<2 in several teleost species. The aim of experiment II is the evaluation of extracellular and intracellular (sarcoplasmic reticulum) calcium sources during the acclimation response to low temperatures using atrial and ventricular strips and ryanodine, a drug that blocks calcium transport from the sarcoplasmic reticulum. In acute studies Hove-Madsen (1992) demonstrated the insensitivity of contraction force at 5rC to ryanodine in the rainbow trout after acute exposure to low temperatures, suggesting that excitationcontraction coupling is exclusively dependent on extracellular calcium sources, what could potentially limit the chronotropic acclimation response.licit Finally, in experiment III the response of the whole organism to acclimation temperature will be analyzed by measuring cardiac output, blood pressure and plasma catecholamines. Any major change found in experiments I and II will be significant for the survival of the individual only if it impairs the ability of the heart to supply oxygen to the tissues. Training content (objective, benefit and expected impact) In the department, intensive interchange of ideas between the trainees and their supervisors is fostered and developed by in several ways. First, the trainees attend a monthly 'physiology journal club', in which newly published papers on cardiovascular or related topics are presented, critiqued and analyzed. Second, there is a weekly departmental seminar program featuring national and international speakers. Third, the supervisor's lab is founded on the principle of extensive daily interaction with other trainees, graduate students and visiting research scientists. The extensive collaboration of the group with laboratories in USA, Canada, Australia, New Zealand and Italy, will stimulate the applicant postdoctoral training as well as potentiate further collaborative research in the comparative cardiovascular field. Links with industry / industrial relevance (22) My project pretends a holistic approach to the problem of temperature acclimation in fish. Aside from the basic importance of understanding cardiac acclimation, there is also a potential short-term use of the results obtained in the field of fish culture. Despite the investments done in the last 20 years, the aquaculture industry is still subjected to important . fluctuations in fish stocks caused by environmental factors, temperature being one of the most important because it is one of the major effectors of seasonal changes. Moreover, temperature changes have an important impact on mortality in fish stocks with a complex etiology but the role of the cardiovascular system has not been investigated. I
