Upgrading of natural products from the Russian Far East for pharmaceutical, cosmetic and food applications

A number of natural products from Far East marine plants and animals and from wastes after industrial processing of crustaceans was isolated and characterized. Carrageenans were isolated from Tichocarpus crinitus and Chondrus armatus, two of the most abundant red algae in the Peter the Great Bay, the Japan Sea. NMR structural investigation revealed that the polymer from T. crinitus was a furcellaran-like polysaccharide with a poor gelling ability and that one from C. armatus was predominantly composed of kappa-carrageenan and showed a gelling ability in the presence of potassium ions. Both galactans exhibited anticoagulant activity. Laminarans were extracted from the brown algae Laminaria cichorioides, L. gurjaovae, and Fucus evanescens. Their structure was investigated by NMR and FTIR spectroscopy, gas liquid chromatography and circular dicroism. The contents of ß 1=>3- and, ß 1=>6-linked glucose residues was determined after enzymatic hydrolysis and HPLC analysis and confirmed by methylation analysis. Molecular weight of the laminarans, determined by HPSEC, were in the range 5000-14000. Effective methods for the isolation and purification of ß 1=>3-glucanase and , ß 1=>6-glucanase from the molluscs Spisula sachaliensis and Chlamys albidus were established. These enzymes were used to enzymically modify laminarans by a transglycosylation reaction, thus obtaining biologically active polymers. Proteases with collagenolytic and elastase activity were isolated from industrial crab processing wastes and characterized with respect to molecular weight, isoelectric point and aminoacid composition. A crab enzymic complex was clinically tested and recommended by the Pharmaceutical Committee of Russia to be used in medicine for enzymatic treatment of burns, osteomyelitis, bedsores, diabetic gangrene. A number of compounds were isolated and characterized by NMR spectroscopy from Eleuterococcus senticosus. The production of such compounds, among which are umbelliferone and a series of lignans, might lead to the use of E. senticosus in a range of applications in medicine and cosmetology.
