Deliverables
This report will include electrical characterization results of a CMOS interface demonstrator, including graphs showing power consumption, noise performance, and transfer function of the recording channels. The report will include characterizations with and without the passive MEA, as well as a description of the MEA/CMOS interconnect.
In vivo LFP detection with conformable passive MEA (>1500 electrodes/cm2) (opens in new window)This report will present photos and characterization graphs (e.g. electrochemical impedance) of the passive microelectrode arrays, as well as time-frequency spectrograms of LFP recordings from a cortical surface with designated behavioural states, high-pass filtered traces of waveforms, trigger averaging of waveforms demonstrating consistent action potential morphology, and autocorrelograms indicative of neural firing refractory periods.
eFUS chip: Biological/histological evidence of efficacy (opens in new window)The report will include the first in vivo results using the eFUS chip manufactured by the consortium. The eFUS chip will be implanted and the animals will recover, and will be stimulated over 5 days according to pre-determined parameters. Histological methods will describe the biological effects of the eFUS (e.g. c-fos, TUNNEL, inflammation markers, etc). Molecular methods will describe regional changes in gene expression in key areas (mPFC, NAc).
Project Handbook (opens in new window)This deliverable will consist of a guide to implementation the UPSIDE project It includes a detailed project management plan including the Gantt chart and the Work Package structure the schedule per task responsible partner for the related subtasks the related deliverables and the dependencies on other tasks The handbook should act as a a reference source for all consortium members covering many daytoday activities
Report on the integration of CMOS 64-channel recording interface with passive MEA (eREC64) (opens in new window)This report will include results of electrical characterization of the CMOS 64-channel recording interface with assembled and interconnected passive MEAs, including: (i) time-domain graphs with the recorded data resulting from stimulation a saline solution with a passive tungsten electrode. (ii) noise plots obtained with a spectrum analyzer will also be reported here, with annotations representing the fully integrated noise in the bandwidth of interest. (iii) transfer-function frequency plot obtained from a network analyzer.
in vivo targeting of FUS stimulation: safety and precision (opens in new window)The report will include the results of in vivo acute studies using commercially available FUS in anaesthetized rats. The data described will concern the safety aspects, and the refinement of the “targeting”. Biological effects of the stimulation will be confirmed during the surgical session using electrophysiological methods, and post-mortem by using histological methods. Regions of interest examined will include mPFC, and NAc, for example.
Plan for dissemination, communication and exploitation (opens in new window)This plan describes the strategy procedures and tool for dissemination communication and exploitation of project results and also focuses on the public engagement
Report on chronic detection of EEG signals for biomarker detection algorithms, in an in vivo animal model with IGT-array for >3 weeks (opens in new window)This report will show photos of IGT-based devices, placement procedure, and EEG data acquired by an IGT array over > 3 weeks period of time. Time-frequency spectrograms of the recordings, high filtered traces of waveforms will be shown indicating potential biomarkers. Spectrograms and raw data will be analysed to understand best methods to extract depression biomarkers.
eFUS chip implantation protocol and preliminary FUS stimulation parameters (opens in new window)The report will include the results of our initial in vivo and ex vivo studies using phantom agar and post-mortem fixed and unfixed brains. These preliminary studies will use commercially available FUS. The report will include protocols and parameters concerning the intensity and targeting of the stimulation which will be transferred over to the device emerging from the other WPs. The report will also describe craniotomy and protocols to implant “dummy” eFUS and eREC chips.
Internal project coordination tool public internet website and other social media channels to communicate and disseminate the project results
This plan will describe what data the project will identify and generate how to handle with it and how the FAIR principles will be implemented
R2 updates on the DMP (opens in new window)First update of the Data Management Plan (DMP)
This report will present electrical characterization results of the manufactured demonstrator of the relay station. This will include time-domain graphs with the input and output signals of a microcontroller used to program and acquired data from the eREC and eFUS devices, graphs with the characterization of the power unit in the relay station and demonstration of wireless connectivity with a computer by sending and receiving programming commands.
Conformable IGT-array (>1500 interfaces/cm2) with superior SNR (>20 dB increase) and >1 month in vitro stability (opens in new window)This report will present photos and characterization graphs (e.g. IVgraphs, temporal response, frequency response) of the IGT array. In addition, we will show comparable acquired electrophysiological signals from IGT and passive arrays with SNR increase. In-vitro stability will be shown with prolonged operation (drain current modulation for pulsed gate voltage) in physiologic conditions.
