Native chemical glycosylation: a novel and efficient method for the synthesis of complex glycoconjugates

Ziel

The glycosylation of proteins is regarded as one of the most complex modification within eukaryotes.
This modification serves to define the biological role of the glycosylated protein in processes such stability and solubility of proteins, immune response, intracellular targeting, intercellular recognition and protein folding. An evident complication of the study of naturally occuring glycoproteins and their role in the before mentioned biological processes is their occurrence as glycoforms. As each component of this microheterogeneous population exhibits different properties, structure-activity relationship studies are not possible.
Besides this, the therapeutic application of non-homogeneous glycoproteins may in the near future be regarded as undesired by the regulatory authorities. With the desire to solve this problem, this proposal presents the development of a novel approach, named Native Chemical Glycosylation, toward the synthesis of well-defined homogeneous glycoproteins.
