Receptor clustering promoted by restricted divalent ligands

Obiettivo

Dr. Trinidad Velasco, a Spanish national who has recently completed her PhD in the UK and been a researcher in Belgium in the area of bioorganic chemistry, is requesting funds for 2 years to research in Ireland with Dr. Paul Murphy and study fundamental aspects of the mechanism of action of multivalent legends. It is proposed that Trinidad will receive training in carbohydrate and oligosaccharide synthesis, application of NMR and molecular modelling techniques in design and determination of solution conformation of divalent legends and in biophysical methods for studying the interactions of the synthesised legends with biological receptors. Collaboration with the group of Hans Joachim Gabions and Sandrine Andre in Munich has recently been agreed. The project will diversify the expertise of the applicant who has worked previously on the area of supramolecular chemistry and molecular recognition of carbohydrates by synthetic macro cyclic receptors. This work provided a first approach to the principles of molecular design and the multiple interactions involved in carbohydrate recognition. The synthesis of a series of macro cyclic polyamide receptors was accomplished and the study of their binding properties both in homogeneous and heterogeneous media was carried out using a variety of techniques, such as NMR and fluorescence spectroscopy titration’s and extraction experiments. The results obtained highlighted the importance of the precise disposition of the binding functionalities and the degree of conformational restriction in the receptors as fundamental factors in order to obtain selective and efficient monosaccharide recognition. During the course of her PhD, completed at the University of Bristol (UK), the applicant gained experience in synthetic organic chemistry procedures and a number of analytical techniques used to investigate supramolecular interactions between the studied hosts and guests.
