Shiga-toxigenic Escherichia coli/infected mammalian cells cross talk: post genomic and molecular targeted approach

Obiettivo

Shiga-toxigenic Escherichia coli (STEG) are highly adapted food-borne intestinal pathogens causing haemorrhagic colitis and the haemolytic uremie syndrome (HUS). STEG constitute a major problem in human food safety as cattle represent an important reservoir of STEC strains. Ability of STEG to adhere to. and subvert normal signalling pathways of, host cells is a key process in their pathogenesis. The objective of this project is to dissect the cross talk between STEC and epithelial cells on both the protein and genomic level. We will focus our study on STEC of serogroup O157 which are the most commonly associated with severe human disease. Host cellular targets of STEC translocated effector proteins will be identified using the yeast two-hybrid technique and their contribution to the integrated process of infection will be assessed. Using DNA micro array we will determine global alterations in bacterial gene expression in response to infection of eukaryotic cells. Available STEC specific oligo chips will allow us to monitor gene expression at different stages of infection and during adhesion to a variety of cell types. These data will deepen our knowledge of the molecular dialogue between STEC and their host cells and could lead to identification of new targets for therapeutics, prevention or controls strategies. Moreover, this project should allow me to considerably improve and complement my previous training in bacterial pathogenicity and will expand my knowledge into novel disciplines of genomics, as well others cutting edge techniques, which will be a strong added value for achievement of my future research career and goals. We also expect that my integration in Pr Frankel's group will lead to strengthen the scientific collaboration and technologies exchanges with my previous lab at the Vet school of Toulouse which also focuses its studies on molecular and cellular basis of E. coli pathogenicity.
