Selective oxidation and cleavage of DNA by photosensitized electron transfer

Obiettivo

Oxidative damage to DNA is of considerable interest because of the key role-played by oxidation in aging, mutagenesis and carcinogenesis. The aim of this project is to help understanding the mechanisms of photooxidation of DNA induced in biological system s by various exo- and/or endogenous photosensitizers.

The experiments will involve the synthesis and photochemical studies of a range of DNA oligonucleotides of specially designed sequences, containing suitable photosenisitizers, namely fluorescent acridi nium or pyridinium cation derivatives as electron acceptors, and guanine residues, as electron donors, at defined positions. Techniques used include solid phase DNA synthesis, photoirradiation, laser flash photolysis, steady-state and time-resolved fluorimetry, high-performance liquid chromatography coupled with mass spectrometry (HPLC-MS).
