MICROBIAL PRODUCTION OF A PRECURSOR (RHAMNOSE) FOR THE SYNTHESIS OF FLAVOURING BY USING A STARCH DERIVATIVE

Ziel

The objective of the project is to obtain rhamnose in a form which permits industrial use of this sugar for the production of aroma and flavour compounds.
The production of natural sugars for use as flavour precursors, and in particular red fruit flavours is being studied.

The process, based on the fermentation of a starch derivative by a selected strain of microorganism, has been developed (m{3} scale) with rhamnose yields of 17 g/l. Downstream processing of rhamnose relies on fermentation and enzymatic extraction and column separation. Industrial scale up (10 m{3} level) is underway.

The project concerns the selection of appropriate strains of microrganisms and the development of fermentation and purification processes. Specifically, the work involves isolation of novel strains from the environment, cultivation of strains on industrial media and rhamnose harvesting. The department isolates and cultivates novel microorganisms and then selects microorganisms and enzymatic separations for the ability to cleave rhamnose from the polysaccharide. We have succeeded in removing protein from the polysaccharide and in recovering rhamnose. Optimization of hydroysis and purification are currently underway.

The project concerns the selection of appropriate strains of microrganisms and the development of fermentation and purification processes. Specifically, the work involves isolation of novel strains from the environment, cultivation of strains on industrial media and rhamnose harvesting. UAB screens microorganisms, develops and optimizes processes for the selective digestion of the polysaccharide for harvesting rhamnose in quantitative yields by separation through use of such industrial processes as continuous column chromatography. UAB has succeeded in developing processes based on selected microorganisms for the obtention of pure rhamnose. Optimization and scale up are currently underway.

Bioeurope isolates, characterizes, selects and cultivates rhamnose producing strains, optimizes fermentation and produces polysaccharide for the development of harvesting and purification processes. Current yields of 35 g polysaccharide pes 1 represents a 20 fold increase accompanied by the halving of fermentation time. Such results were achieved through optimization of the fermentation process, including media, inoculation, pH, temperature, aeration and agitation.
The work programme involves strain selection, cultivation, and the optimization and scale up of production and the selective extraction of rhamnose.

The results will include the selection of a microbial species forming a polysaccharide containing high amounts of rhamnose and the selective release of rhamnose through enzymatic, fermentative, or chemical hydrolysis. Novel microbes for use in extraction will be developed. Applications in the natural aroma area will be developed.
