Improvement of lactic acid bacteria for traditional and novel applications in biotechnology

Objectif

Lactic Acid Bacteria (LAB) are industrially important in a range of food, drink and feed industries indigenous to Europe and in addition, their positive roles in human and animal health are receiving increased attention. The G-Project on LAB is organized as a single integrated project that combines the expertise of 36 participating laboratories. The Project is divided into sub-groups to facilitate a targeted focus on key objectives. The sub-groups are: gene regulation; bacteriophage (phage) and phage resistance; bacteriocins; LAB in beer and vaccines.
The project has made impressive progress since its initiation in December 1994. It is clear that the knowledge base and analytical techniques have become sufficiently advanced to address questions regarding the expression of key technologically important traits of LAB. Thus, not only has the organisation of the genetic loci for a range of significant traits such as bacteriophage resistance (abortive infection, restriction/modification), bacteriocin production and immunity, and amino acid biosynthesis been elucidated but the complex networks by which these activities are regulated are also being unravelled. The work on bacteriophages of LAB is yielding relevant information on phage/host interactions which is a key element in generating strategies for combating phage infection in commercial practice. The fundamental and applied research on bacteriocins has highlighted features regarding induction of bacteriocin activity, the role of media constituents in affecting activity spectrum and the successful application of some bacteriocins in food trials. Potentially new taxa of beer spoilage LAB have been identified and detection methods are being refined and made more sensitive. A significant advance has been the demonstration of the feasibility of using LAB as presentation hosts for foreign antigens in order to induce a host immune response. These advances stem from an excellent integration of the disciplines of Microbiology, Molecular Biology and Immunology.
