Individual radiosensitivity: heritability and genetics

Obiettivo

Clinical data from radiotherapy and experimental evidence obtained in vitro demonstrate the existence of inter-individual variation in response to ionising irradiation. The contribution of age, genetic polymorphism and environmental factors to these variations is still unknown. We propose to identify cellular phenotypes, related to individual radiosensitivity. This will permit the identification of involved functional and positional candidate genes, whose polymorphisms actually modulate individual radiosensitivity. The proposed approach is based on the exploitation of the unique GENEPI biological resource, the use of two recently validated, quantitative in vitro assays of radiosensitivity, and on the application of state-of-the-art gene expression profiling technology. Results of the study are intended to lead to the development of a predictive test for individual radiosensitivity, capable of identifying persons at risk.
