Obiettivo
Research objectives and content Our aim is to develop immunoaffinity columns to apply in the extraction of various types of samples that contain taxanes. Polyclonal antibodies against taxanes, will be isolated from animal and will be immobilised on an appropriate support. The selectivity of the immobilised antibodies on the recognition of taxanes will benefit on the selective isolation of the compounds of interest from complex extracts. Another advantage of the proposed method is the sensitivity of the immunoassay. Samples having low taxane content could be extracted without the problems phased by conventional SPE. and subsequently analysed by HPLC-diode array with the methods we have already developed. HPLC analysis could be simplified and probably prove more informative since numerous contaminants would already be discarded during the immunoassay. By combining the selectivity of the immunoassay with the chromatographic results (retention and spectral data) the confirmation of the nature of the analytes would be easier, thus reducing the need of using LC-MS analysis. Training content (objective, benefit and expected impact) The main benefit obtained from the requested training will be the involvement of the applicant with a new discipline and methodology. Apart from that, the initiation of a new project in the Laboratory of Analytical Chemistry AUTH will profit to the institutions as well. A new technique will be established in the laboratory and the transfer of expertise and knowledge on this topic from the co-operative laboratories will benefit not only to the applicant but also to the host institution. The requested project can work as the first step in a future closer collaboration of the interrelated laboratories. Links with industry / industrial relevance (22) The production of taxol and related compounds has a great industrial and economic interest. Due to the low yield of taxol from extraction of yew bark and restrictions on the harvest of yew trees, alternative sources like cell suspension cultures have to be found. The knowledge acquired from the proposed project can improve the systematic analysis and screening of plant or cell culture samples, thus contributing to the selection of high producing varieties or cultures.
