Rapid determination of organic microcontaminants in the water/sedimentinterface layer using fast extraction procedures with subsequent gc- - tandem-ms analysis

Research objectives and content
The increasing concern about the environment and its pollution has led to the development of analytical techniques for the determination of organic micropollutants at adequate levels desired by, e. g. EU directives. However, the demand for trace-level analyte determination in the various compartments in the environment, and the growing amount of samples, require a novel strategies such integrated sample handling and chromatographic techniques. The need for unambiguous structural identification request very sophisticated detection. In the study proposed here, analytical systems will be optimized and applied to samples rather from the top layer of river sediments. This part of the aquatic is especially interesting, because (i) the concentration c even polar and good-water soluble analytes are commonly much higher than in the water (which is more frequently analyzed), (ii) it hosts many biota (e.g. worms, plants, fish; but also microorganisms) which makes it a region for bioaccumulation, biodegradation as long as bioavailability is present, and (iii) scientific interest (analyte concentration; distribution between water and solid compartments). The analytical system will be based on the on-line/at-line combination of a sample preparation modules, i.e. solid-phase extraction for water while in-vial extraction and MASE are used for water and solid sample The combination of integrated sample preparation with gas chromatography with sensitive and selective MS, MS/MS and AED detection, will permit a sample reduction from typically 40 g to less than lg. This will require optimization of large-volume techniques. The practicality of the proposed procedures will be demonstrated for different types of real-life sediments. Target analytes are mainly (organo-sulphur, phosphorus, organonitrogen, -halogene and -tin compounds. Both target analysis and identification of unknowns will be persecued. Emphasis will be given on microcontaminants typical for the regions of both host institute and candidate.
Training content (objective, benefit and expected impact)
During the period of 18 months several new analytical techniques will be used by the candidate. Regarding sample treatment; MASE will be trained in the laboratory of dr. Dean at the University of Northumbria in Newcastle (United Kingdom). The present knowledge on AED and MS/MS is insufficient and needs to be updated. Working with the proposed sample handling and detection techniques combined with the present understanding will greatly enlarge the candidates analytical view.
