Extreme conditions for studying protein stability and regulation

Objective



In the last decade an increasing interest of researchers from both fundamental and applied domains has been focused on studying protein stability and enzyme regulation under extreme conditions. Together with the attempts at better understanding the proteins from extremophiles (thermophiles, halophiles, barophiles etc.), extreme conditions are used for the enzymes from mesophilic sources. The goal of this project is two-fold: determine structural principles lying at the basis of function and stability of proteins and more complex systems; and adapt the enzymes to extreme conditions of high temperatures, high pressures, and organic solvents.

The practical objective of such adaptation is obvious: a vast amount of processes in the food and pharmaceutical industries, fine organic synthesis, analysis, and other biotechnological areas demand short-term or prolonged exposure of proteins to what is called extreme conditions. In this case, care should be taken in order to prevent protein deterioration and success can be achieved based only on a knowledge of protein behaviour under extreme conditions.

Special interest will be paid to denaturation of enzymes by temperature, high pressures, and organic solvents with the aim of eluciding some common features responsible for protein stability. The possibilities of regulating the enzyme activity and specificity under extreme conditions will be studied.
