Novel cis- and trans-elements involved in transcriptional control in eukaryotes

Obiettivo



In most eukaryotic model systems the current knowledge of transcription control is limited to the interactions between regulatory proteins bound to enhancers (or silencers) and the basic transcription machinery bound to promoters. However, it appears that in vivo several further factors (different auxiliary proteins, proteins determining long distance interactions, proteins interacting with insulators, etc.) are involved in the process.

This project bring together three leading European institutes of molecular biology for the study of novel types of cis- and trans- regulatory elements involved in transcription control. The project will start in Moscow with the cloning of recently discovered Drosophila regulatory elements necessary for interactions between sequences separated by distances of up to 1-2 megabases as well as the cloning of three genes, e(y)1, e(y)2 and e(y)3, responsible for long distance interactions in D. melanogaster. In parallel with the structural analyses, the functional analyses of these elements of the second phase will be initiated by the groups in Heidelberg using an established Drosophila-based cell free system.

The Strasbourg group will provide technical suggestions in the first phase and will contribute from the second phase onwards by screening for mammalian homologues of the Drosophila factors.
