MULTIDISCIPLINARY ANALYSIS OF THE MECHANISM OF CARDIAC EXCITATION- CONTRACTION COUPLING USING THE PRINCIPLE OF TRANSIENT PERTUBATION

Obiettivo



Studies of cardiac excitation contraction coupling will be conducted in a collaboration between a laboratory studying isolated myocytes (Essen), one studying papillary muscles, and subsequently isolated myocytes (Lund) and one studying papillary muscles, intact dogs and humans (London). In each preparation the effects of transient pertubations of beats within a steady train will be studed, i.e. premature excitations, single prolonged depolarisations, and the potentiation and decay which follow these. Interrelationships between systolic and diastolic membrane potential, second inward current, and contractility will be analysed in order to attempt to distinguish the roles of second inward curent, sarcolemmal calcium binding and release, and sodium/calcium exchange,in calcium induced calcium release from the sarcplasmic reticulum, and the regulation of the amount of released calcium.
