Cel The objective of the proposal is to use cytogenetic methods to examine fundamental aspects of variations in radiosensitivity between individuals. The proposal exploits recent advances in using the G2 assay in lymphocytes to identify normally responding and hypersensitive individuals amongst cohorts of healthy subjects and cancer patients. Current evidence suggests that G2 sensitivity is a marker of predisposition to cancer. Suitable normal and sensitive subjects will have small skin biopsiestaken and from these fibroblast cultures will be initiated. These will provide a stock of cells that will be irradiated in vi tro and examined for induced chromosomal damage. Particular emphasis will be placed on the presence of stable aberrations detected by using the fluorescence in situ hybridisation method on mitotic preparations. The data will be examined for both quantitative and qualitative differences in chromosomal responses between individuals that may correlate with their known radiosensitivity and disease status.
