A subset of cultivars, common to all collections will be considered for the following aspects: tree shape and behavior, variation on oil acidity composition and polyphenols content, abiotic stress tolerance and fruit production.
Capacity of main olive varieties and wild plants to face drought stressAll information available on the potential to grow under drought stress conditions will be organized into the common database.
Capacity of main olive varieties and wild plants to face salt stressAll information available on the potential to grow under salt stress conditions will be organized into the common database.
Common protocol to collect morphological parametersA common protocol to collect morphological parameters will be established. Aspects to consider include the number and type of traits, phenology, representative samples, imaging technology, softwares for data elaboration and data output.
Identification of the most relevant aspects to explore by association mapping strategiesMain traits that will be explored include: oil quantity, polyphenols content, acidity composition, tree habit, fruit size, pulp/stone ratio, harvesting season, drought and cold tolerance, diseases (Spilocaea oleagina, Pseudomonas savastanoi, Verticillium dahliae, etc). Markers linked to these traits will be searched by the approach of association mapping.
Phenotypic profile of varieties related to tree vigour and canopy shapeAll information available on tree vigor and canopy shape will be organized into the common database.
Inter-incompatibility for the most important 200 varietiesData made available on inter-incompatibility of cultivars will be organized into the common database.
Development of new cytoplasmic (plastidial and mitochondrial) markers able to discriminate cultivars, wild plants and related subspeciesA new set of chloroplast markers will be developed starting from the set already published, including those able to discriminate among wild plants and related subspecies. A new set of cytoplasmic markers will be also made available from the whole mitochondrial genome sequence.
Phenotypic profile of varieties related to fatty acid olive oil compositionData available on oil fatty acid composition will be collected for each cultivar and will be compared among the main collections considering other aspects such as: area of diffusion, altitude, latitude and climatic conditions, etc.
Capacity of main olive varieties and wild plants to face heat stressAll information available on the potential to grow under heat stress conditions will be organized into the common database.
Rules concerning the access rightsLRU will Identify the legal and ethical problems linked of the construction of the databank. In particular the problems as follows will be taken into consideration: - Problems concerning the property and the other rights relating the databank and the genetic material; - Problems related to the access rights.
Development of new polynucleotide-SSR markers able to discriminate among cultivarsA selection based on their discrimination power of poly-nucleotide SSR as tri-, tetra- and penta-nucleotide repeats will be done, from the whole genome sequence and from the numerous RNA libraries made available by partners
Best varieties associations for the most relevant olive producing areasAll information available on the best association of cultivars related to climatic, soil and biotic/abiotic stress resistance for the most relevant olive producing areas will be organized into the common database.
Guidelines on the legal and ethical management of the germplasm databanks SECOND RELEASEUpdated version at the end of the project
Phenotypic profile of varieties related to the early bearingAll information available on early bearing will be organized into the common database.
Core list of varieties (about 200) representing the main olive oil producing countriesA list of the most relevant varieties contributing to the olive oil production for the Mediterranean countries and new areas of olive cultivation of South (Argentina and Chile) and North America (Mexico) will be provided. Reference information will derive from International, National and Regional Olive Varietal collections previously characterized by molecular and morphological markers.
Molecular profiles of olive varieties previously characterized for agronomical/phenotypical characterMarkers linked to traits of agronomical, morphological or biological interest will also be searched in order to assign a specific trait to a relative marker
Self-incompatibility for the most important 200 varietiesData made available on self-incompatibility of cultivars will be organized into the common database.
Phenotypic profile of varieties (resistant/susceptible) related to the olive leaf spot (Spilocaea oleagina) diseaseData available for each cultivar on susceptibility to olive leaf spot will be compared among the main collections considering other aspects such as: area of diffusion, climatic conditions, pathogen strain, etc.
Development of new SNP markers able to discriminate among cultivarsThe best SNP (Single Nucleotide Polymorphisms) markers, developed by the partners of the project by using Next Generation Sequencing (NGS) approach, will be selected to establish a common set of markers for cultivar discrimination and to build a SNP array.
Awareness and Dissemination planAn awareness and dissemination plan will be elaborated and will be updated and deployed along the project life and beyond. The different versions of the Dissemination Plan will include the activities that have actually been developed in the project in the previous plan and those foreseen for the next period. The Dissemination Plan will contain the following elements: 1) Identification and classification of target stakeholders to be addressed. 2) The dissemination methods and their specific associated activities. 3) Schedule and complementarily of the dissemination activities among partners. 4) The conditions to ensure proper dissemination of the generated knowledge. As was previously commented, the identification and communication with stakeholders is of main importance. The first Awareness and Dissemination Plan will include a list of stakeholders and the plan to address those, a close contact must be established, offering information in different formats in order to ensure it adequacy to the needs of each group of interest. This kind of contact will start early in the life of the project
Phenotypic profile of varieties (resistant/susceptible) related to the olive fruit fly (Bactrocera oleae) insectData available for each cultivar on susceptibility to olive fruit fly will be compared among the main collections considering other aspects such as: insect diffusion, number of generations, meteorological parameters, level of damage, etc.
Final release of a common set of molecular profiles of the olive cultivars, wild populations and related subspeciesThe same action, previously described, will include also wild plants and related subspecies discriminated by a specific marker set.
Phenotypic profile of varieties (resistant/susceptible) related to the Verticillium (Verticillium dahliae) diseaseData available for each cultivar on susceptibility to Verticillium wilt will be compared among the main collections considering other aspects such as: area of diffusion, irrigation, tree damages, etc.
Phenotypic profile of varieties related to biophenols, secoiridoids, steroids and tocoferols olive oil compositionData on oil composition in biophenols, secoiridoids, steroids and tocoferols will be reported for each cultivar on a common database.
Economical and Institutional aspects second releaseA second and final release of the economical and institutional aspects at the end of the analysis.
First release of a common set of molecular profiles of the olive cultivarsFrom the analysis of 200 main cultivars with the set of markers selected within this project it will be possible to obtain a unique and stable profile for each variety, accepted as reference profile at world level.
Phenotypic profile of varieties related to the varietal useAll other information available on the varietal use, such as table olive varieties, oil characteristics, varieties showing strong adaptation to extreme conditions, will be organized into the common database..
Seminars and Workshops will be organized in correspondence of each deliverable.
First release of a molecular profiles database for olive cultivarsA common database including all cultivar profiles will be built, able to compare olive varieties for their identification and discrimination.

Final release of a common molecular database for olive cultivarsThe final Database version including the molecular profiles of cultivars, wild plants and related subspecies will be definitively published.
Public WebsiteProject website “BeFOre” project will have its own web page, developed and maintained by ISAFOM. It will be used for the internal communication, and it will also form the basis for the administrative and the external communication. A book reporting the main results will be published on the web site of the project; Internal information will be restricted to partners, which have different rights for viewing parts of the information structure. The web site will also be used as a basic tool for external communications. In the public area we will develop a communication strategy towards the general public and selected target group. Here we will collect all material, which should be publicly accessible (presentations, papers, information material, posters, etc).
Establishment of a highthroughput platform for olive cultivars molecular characterizationBased on the markers developed, a common platform will be established to discriminate among olive cultivars, wild plants and related subspecies in each lab involved in the project.
Final release and online publication of a common database on morphological parametersThe final release of a common database will be done as an online free access version and as publications.
