DEVELOPMENT, VALIDATION AND APPLICATION OF MOLECULAR MORPHOLOGICAL AND PHYSIOLOGICAL MARKERS FOR JUVENILE TO MATURE STATE CHARACTERIZATION IN WOODY PLANT SPECIES

Obiettivo



The specific objectives of this research project are: The identification Of markers differentially expressed in juvenile versus mature phases of tree species which include: the generation and characterization of phase specific CDNA markers by means of differentially expressed genes. the analysis of anti-oxydant isozyme patterns related to phase change the analyses of changes in meristem profile and zonation during aging. the characterization of developmental stages by in vivo monitoring of cytoplasmic pH, changes in membrane potential and ion distribution. the determination of specific plant growth regulator ratios and signaling to characterize maturation and rejuvenation.
The testing of such markers for specificity across a range of species (Gymnosperms and Angiosperms) and tissues (Juvenile and mature) to determine their general applicability.
The performance of field trials at larger scale for validation.
During this project different types of markers for phase change from juvenile to mature stage will be identified in certain model species (peach, apple, pines,...) and developed for different forest species (Pinus sp., Prunus avium). These markers include molecular markers based on tissue specific mRNAs, isozymes (CAT, SOD), plant growth regulator ratios (Auxins, Cytokinins, ABA, Ethylene,PAs, Phenols, Ion content) and other physiological, histological and morphological markers. In order to test their specificity and general applicability, molecular markers developed by individual participants will be interchanged and tested by other groups on different plant species and using different juvenile, mature and rejuvenated tissues (e.g. shoots, nodes, buds, phytomers; seedlings and young progeny genotypes, cascade graftings, cuttings, micrograftings, micropropagated material). On the other hand, plant material of different species will be made available to the participants studying isozymes, plant growth regulators, physiological and histo-morphological markers in order to extent their analyses to other species and tissues. Plant material with known favourable and unfavourable propagation behaviour will be analyzed with previously established markers. Propagation and flowering behaviour, as predicted by marker analyses, will be verified at larger commercial scale through field trials.
Phase specific markers would offer the possibility to identify genotypes with prolonged Juvenile phase, to evaluate any process for rejuvenation, or to identify genotypes with early flowering for seed orchards. In this way powerfools tools to manipulate propagation (afforestation) and for tree selection and breeding would be available.
