Analysis of the transferrin gene expression in the central nervous system and its role in myelinogenesis

Research objectives and content
Deficiencies in myelin lead to grave disorders in the nervous system. It was observed that variations in transterrin (Tf) gene expression in oligodendrocytes of the central nervous system (CNS) may have important consequences in myelination process. The aim of the work is to understand the mechanisms responsible for the Tf tissue-specific expression in the CNS, as well as to understand the Tf role in the CNS development, particularly in oligodendrocytes development and in its influence on myelin synthesis. For this purpose, ex vivo analysis of Tf gene expression will be performed with oligodendroglial cell lines in presence or absence of various physiological effectors and using culture conditions described for the induction of oligofrndroglial l differenciation. In addition, analysis of Tf gene expression will be studied in vivo in the CNS of transgenic mice overexpressing human Tf. The expression of some proteins markers of myelin will be also analyzed in cultured oligodendrocytes as well as in transgenic animals. Obtention and analysis of transgenic animals, cell cultures, RT-PCR, transient expression experiments in l cultured cells, in situ hybridization studies and DNA-protein interaction analysis will be the methods used.
Training content (objective, benefit and expected impact)
To obtain a PhD. in University Paris Vl in France. The thesis work will give me the necessary tools to continue my scienitic carrier under the best conditions.
