Obiettivo
We propose to develop a microfluidic chip-sized assay for low-cost, sensitive, rapid, and multiplexed detection of bacteria in urine. We aim to reduce bacterial diagnostic time from the current standard of 1-3 days to several minutes. The device will have application to both conventional clinical microbiology laboratories and point-of-care settings, and will eliminate dependence on highly trained operators. We propose to use isotachophoresis (ITP) to extract and concentrate the bacterial signature 16S ribosomal RNA directly from urine samples, and detect and quantify its presence using a set of complementary fluorescent probes (molecular beacons) on a low-cost microfluidic chip.
