Mechanisms of resistance in candida species in aids

1) Training in the methodology of antifungal susceptibility testing. The applicant would be taught how to perform in vitro susceptibility testing of Candida species. The applicant will assess in vitro the activity of a novel antifungal compound in collaboration with an industrial partner and participate in work on the validation of in vitro susceptibility testing in the clinical setting.
2) Training in real-time imaging microfluorimetry. The applicant will learn how to study changes in intracellular pH and [Ca2+] using imaging microfluorimetry. He will determine whether azole resistant isolates of Candida exposed to an azole show a more profound rise in intracellular pH or calcium compared with azole susceptible isolates. Furthermore, he will examine the pHi and Ca2+ changes in pairs of susceptible and resistant clinical isolates to fluconazole.
3) Training in azole antifungal population pharmacokinetics. Using a population pharmacokinetics approach, the applicant will determine whether late stage AIDS patients have equivalent serum concentrations of fluconazole as those with higher CD4 counts. Serum concentrations will be measured by bioassay and the applicant would learn this simple procedure.
