NMR structures of membrane proteins, complexes and lipid assemblies; a dedicated wide-bore ultra-high-field MAS NMR spectrometer for biological research

Exploitable results

Features: - To demonstrate the construction and the use of a novel ultra high field (750 MHz) Magic Angle Spinning NMR spectrometer. - To establish the first MAS NMR facility for structural biology in the world. - To establish and demonstrate crucial improvements of long-term instrument stability range, resolution and sensitivity of MAS NMR technology. - To validate novel concepts for structural and structure-function studies. - To transfer knowledge from MAS NMR technology producers to users. Benefits: - The prototype 750 MHz wide bore spectrometer has become operational on schedule and installation of the instrument is planned for in the second half of 1999 in Leiden. Several probes are operational and the construction of more probes proceeds on schedule. - Novel concepts for structure determination based on heteronuclear correlation spectroscopy and Magic Angle (Oriented) sample spinning are continuously being worked out. - 13C,15N isotope labelling of the histamine ligand and a model protein, the SH3 spectrin signal transduction domain, was realized. Specification of ultra high field performance on these models is now in progress. Membrane protein models have been prepared. - Software for acquisition and processing is currently being improved in a continuous process with partners travelling to the manufacturers site to work with the novel spectrometer ant to provide feedback to the manufacturer. Simulation of NMR data was improved and tested between partners. - An extended audience has been implemented. Dissemination is currently augmented through participation of partners in a broad range of meetings. The most important benefit at this stage is the impact of the project in the biological MAS NMR community worldwide, our first extended audience. The project played a seminal role in orienting the biological MAS NMR towards higher fields. Several European countries are now planning for similar centres as the prototype facility in our project. Although most of these are still in a planning stage at present, we expect soon to see several of these being selected for realization. The European NMR manufacturing industry will be a primary beneficiary in this newly developing market, commercializing what may be considered a unique example of European high-tech.
