Research objectives and content
The objective of this proposal is to produce large quantities of small oligopeptides in a strain(s) of the lactic acid bacterium, Lactococcus lactis. A particular strain of L. Iactis, shown under certain conditions accumulate intact oligopeptides intracellularly, is available in the host institute. This strain will be further manipulated to remove two additional endopeptidase activities. The strain(s) will subsequently be used to express synthetic genes encoding for the oligopeptides met-enkephalin, bradykinin, and a-casein fragment f(1-23). Genetic and physiological parameters will be examined to optimize levels of gene expression and oligopeptide production and purification. Training content (objective, benefit and expected impact)
The training content of this project would offer an expert (the applicant) in biochemical techniques involving protein/peptide purification and characterization the opportunity to obtain expertise in molecular biology. Therefore, this project would allow for the integration of expertise of both the applicant and the host institute. Links with industry / industrial relevance (22)
The proposal is of pre-competitive nature. It will allow the possibilities of producing oligopeptides directly in bacteria to be ascertained. Successful production of oligopeptides using a specific strain developed in this proposal could be of significant industrial relevance. This project could allow the development of an innovative and economically viable alternative to existing methods of peptide synthesis. The proposed method has the potential to efficiently and accurately produce oligopeptides in large quantities and may be superior to existing chemical synthesis or gene fusion methods.
