Identification and analysis of genes required for early oogenesis in 'drosophila melanogaster'


Research objectives and content
The establishment of polarity and patterning in development are initiated during oogenesis. Oogenesis in the fruit fly Drosophila melanogaster has been studied genetically by the isolation of mutations which cause females to be sterile. This approach does not allow the identification of genes which have an essential function at other points in development. I will perform a genetic screen specifically aimed at isolating this second class of genes. With this aim, I will use heterozygous flies and study the effects of the mutation in oogenesis in clones homozygous for the mutations. The clones will be generated by mitotic recombination, and will be marked by the expression of a fluorescent reporter. I will dissect the ovaries of the flies, identify the homozygous clones by the expression the marker and analyse their phenotype. If oogenesis is defective in the clones, I will establish lines of flies carrying the mutation, which will be studied further.
The screen will complement other genetic approaches
previously used to the study of the oogenesis. It will allow the identification of genes involved in oogenesis, which will be then amenable to molecular analysis.
Training content (objective, benefit and expected impact)
Carrying out the research project described above will allow me to become familiar with a variety of genetical and cell biological techniques applied to the fly Drosophila melanogaster. This organism is an excellent model for the study of problems in cell biology and development, and acquaintance with these methods will be very useful for my career. In addition, the research environment in the laboratory and in the Development Programme at EMBL will allow me to acquire a solid background in developmental biology.
