Identification of genes implied in the excysment process of the hipotrich ciliate oxytricha

Research objectives and content
Under adverse conditions, the ciliate hypotrich Oxytricha forms kinetosome-resorbing cysts, in which the nuclear activity is low. When favorable conditions comes the nuclear activity starts and cell begin, the excystment process. In Oxytricha this process have been well established at the structural level but not at the molecular one. The purpose of this research project will be to acquire molecular information' at the onset and during the course of the excystment process. To do this the genes activated in the' excystment stages will be isolated and sequenced. I will focused in those excystment-activated gene codifying for proteins that either regulate transcription or participate in the acquisition of cell morphology.
Training content (objective, benefit and expected impact)
The objectives are: (i) to construct a catalogue of Oxytricha genes that are transcriptionally active during the excystment process; (ii) to determine the precise cascade of genes being trancribed from the start tothe end of excystment. This objectives will be achieved using common molecular biology techniques, a library construction and screening, cloning or PCR. The use of those sophisticated techniques will benefit my training, as well as the scientific and discussion atmosphere of a multidisciplinary laboratory such as the host institution.
