Elemental speciation analysis in biomolecules

Cel

The present proposal represents an effort in the search for new tools for the speciation analysis of bio-inorganic molecules (BIM), particularly containing Se and Hg. The project is aimed at the evaluation of the importance of Se and Hg-containing proteins in toxicological responses to essential (Se) and toxic (Hg) elements as well as the interferences due to heavy metals exposure on the metabolism of essential elements in the human body. This will be achieved by the development of new multidimensional analytical approaches that allow the identification, characterization and determination of the species involved with a special emphasis on BIM.

The project will be carried out by interfacing high performance liquid chromatography (HPLC) with element specific detectors such as inductively coupled plasma mass spectrometry (ICP-MS). The use of both Sector Field and Quadrupole based mass filters will be exploited. Several approaches, including speciated isotope dilution mass spectrometric (SIDMS) techniques for the accurate determination of the species and electrospray (ES-MS) mass spectrometry for the identification and characterization of the protein content will be developed and applied. The correlation of the speciation results, with the level of metal exposure will be used to study the possible implications of the presence of metalloproteins for biomonitoring and toxicological purposes in humans.

The integration of approaches typically used in molecular biology for the speciation studies by coupling them to elemental mass spectrometry will allow a better elucidation of the role of the BIMspecies identified by the use of multidimensional analytical techniques in the various and "compartments" of the human body.
