New silicon-derived environmentally-benign Lewis-acid catalysts for asymmetric synthesis

Cel

Lewis acids-promoted reactions of organic compounds have been extensively studied in recent years. Many region-, diastereo-and enantioselective reactions catalyzed by a wide variety of Lewis acids have been reported. Still they often have major drawbacks when applied to large scale synthesis: (1) use of toxic metal-derived catalysts, (2) low turn-over, (3) formation of large amounts of salts, (4) low chemoselectivity when applied to polyfunctional reactants, (5) extensive use of control elements ( activating or protecting groups), (6) use of significant amounts of solvents. A few years ago a new class of silicon-derived superacids ( trialkylsilyl triflimides) has been discovered by the group of Ghosez. These environmentally-friendly catalysts have received much attention in recent years.
Our project is (1) to prepare a new class of tuneable silicon-derived “green” catalysts, (2) to test the catalytic activity of these new Lewis acids in representative reactions ( Diels-Alder, aldol, benzylation, allylation, Friedel-Crafts alkylation), (3) to prepare chiral catalysts (4) to test these catalyst in enantioselective syntheses.
The candidate has a background in synthesis of bioactive compounds. He is now completing his training at the European Institute of Chemistry and Biology(IECB) in Pessac in the group of Prof. L. Ghosez. The training programme has been designed to ensure that the candidate’s ambition to acquire a broad formation in synthetic chemistry and pursue a career in that field will be fulfilled. Presently he works on a project supported by industry and is being trained in total synthesis of natural products analogs using original approaches which will required the development of new environmentally friendly catalysts.
Training will be provided by the host group Ghosez at IECB. Additionally the candidate will benefit from the multidisciplinary character of the Institute where organic chemists, physical chemists and biologists interact everyday.
