We will setup two melanoma mouse models the transplantable B16F1 in C57Bl6N mice and spontaneous HGFCDK4 models for which the phenotype and transcriptomic profile of antitumor T cells have been characterised Tumor cells will be injected subcutaneously and stem cell memory potent antitumor T cells or effector memory dysfunctional antitumor T cells Pmel1 transgenic T cells will be injected intravenously 7days post tumor take
Tumor size will be measured daily over a tendays period after which tumors will be harvested and analyzed by multiplex immunohistochemistry coupled to highdimensional flow cytometry to define the 3D spatial distribution of fibroblastic stromal and immune cells and the extracellular matrix and to quantify their number variation and phenotype over time in the tumor microenvironment and draining lymph nodes
At project start BCC will create a housestyle to be used to set up the project website and create a project summary in text and visuals Every partner commits to writing 2 blogposts per year according to the Dissemination and Exploitation Plan ensuring a good timing and relevant spread of result posts throughout the project The website text and visuals will be transferred to a regularly updated slidedeck for project presentations A project brochure will be generated that includes project outcomes to provide stakeholders with an overview of the work done and to generate impact after the project
