Cel Within this project research will be performed at KREATECH Biotechnology B.V ., at the Department of Molecular Cell Biology of the Leiden University Medical Center (LUMC), and at the Department of Applied Physics of the Delft University of Technology (TU Delft). All laboratories are (scientifically) highly skilled, modem, and with state-of-the-art equipment. KREATECH is a biotechnology industrial organization with patents on the Universal Linkage System (ULS^R) and Mycobacterium diagnostics (ABC). KREATECH has broad knowledge of the biochemical and biological properties of ULS^R and on the development and production of biochemicals and diagnostical based on these patents. LUMC is highly skilled and experienced (amongst others) in the in-vitro (molecular) detection of biomolecules and (more recently) in the development of micro array technology. The TU Delft is highly skilled and experienced (amongst others) in the development of (nano-litre scale) biosensor technology. All institutes are (inter)nationally highly acknowledged and have extended (scientific) networks, both national and international. Moreover, all institutes have vast experience in guiding and teaching fellows at all scientific levels. In this project research is proposed to significantly increase diagnostic assay sensitivity up to a factor of 100.000 (10^5), both for nucleic acids and proteins. This problem will be tackled at:1) the level of the detection format,1a) by miniaturization ('from ELISA/dot blot- via micro array- to biosensor format'), and1b) by optimization of biomolecule - ULS^R- solid support binding, and2) the level of sample processing (by a direct ULS^R 'target labeling' technology).
